Dynamic Mitochondrial Networks in Cancer

Research projects evolve in a fortuitous manner, often guided by a convergence of novel observations, intuition, helpful colleagues and unique personal circumstances. It is precisely this constellation that prompted two cardiologists to study the mitochondrial networks in lung cancer cells.

In 2008, my colleague and friend Stephen Archer, a Professor of Medicine at the University of Chicago, asked me whether I would be interested in studying the role of mitochondrial networks in lung cancer cells. My first response was the question ?Do mitochondria really form networks??, because at that time the expression ?mitochondria? evoked images of scattered oval-like organelles, a textbook image of electron microscopy.

I was also intrigued by my colleague?s request, since we were both cardiologists and it therefore appeared to be somewhat unusual for us to study cancer cells. However, as is often the case in science research, personal motivations lay behind Stephen?s newfound research interest ? Stephen?s cousin had recently died from lung cancer.

His cousin?s untimely death and Stephen?s frustration at the lack of therapeutic options for lung cancer victims had served as an incentive for him to expand his ongoing work on the role of mitochondria in cardiovascular cells to also include the investigation of mitochondria in cancer cells. Could we contribute to the identification of novel approaches to treat lung cancer?

During the preceding years, Stephen had focused on the role of glucose oxidation in cancer, in part inspired by the work of the German Nobel prize laureate Otto Heinrich Warburg (1883-1970). In the 1920s, Warburg hypothesized that cancer cells primarily rely on non-oxidative glycolysis instead of glucose oxidation to fuel their energy demands.This metabolic signature of cancer cells was critical for the development and growth of tumors.

As he examined the metabolism of malignant lung cancer cells and non-malignant healthy epithelial cells, Stephen had noticed an important difference in the physical appearance of the mitochondria. The mitochondria in the vast majority of cancer cells appeared to be small and fragmented, while healthy epithelial cells predominantly contained elongated, filamentous-like mitochondria that formed large intact networks. The cause and significance of this difference in the mitochondrial structure between lung cancer cells and healthy lung epithelial cells was unknown and thus a fertile ground for new discoveries.

Even though the planned collaborative project would primarily focus on the mitochondrial network structure and not the mitochondrial metabolism of cancer cells, I also decided to read some of the original Warburg papers in the original German language. German used to be a major language of scientific communication and publication in the 19th century as well as the first half of the 20th century. However, during the latter half of the 20th century and especially in the 21st century, English has become the predominant language of the scientific enterprise, even in Germany.

My nostalgic longing for reading scientific articles in German and my curiosity about how scientists wrote articles in the 1920s prompted me to download some of the Warburg papers. I have to admit that I was quite impressed by the comprehensive nature of the work described. The paper entitled ??ber den Stoffwechsel der Carcinomzelle? (Biochemische Zeitschrift 152, 309-344 (1924)) by Warburg and his co-authors Karl Posener and Erwin Negelein contains a comprehensive evaluation of the respiration of tissues from multiple organs, such as the epithelium, connective tissue, brain tissue, retina and various benign and malignant tumors. This 36 page paper includes numerous hypotheses, observations and conclusions about the nature of tumor metabolism that would inspire subsequent generations of scientists.

One observation made by Warburg, for example, toward the end of the manuscript is that tumors with high levels of glycolysis are also associated with high levels of ammonia production and Warburg refers to this observation as an oddity that needs further research. It would take at least 80 years for researchers to understand some of the key underlying molecular mechanisms that explain this observation, when multiple research groups demonstrated that cancer cells use the amino acid glutamine as a major mitochondrial substrate and which upon degradation releases ammonia.

After reading the awe-inspiring Warburg papers, I felt even more enthusiastic about embarking on this new collaboration to study mitochondrial networks in cancer cells.

Peter Toth, a pharmacologist and neuroscientist who directed the confocal microscopy imaging core in Stephen?s group, used his extraordinary live-cell imaging expertise to visualize the mitochondrial networks of malignant and non-malignant lung cells over time. The microscopy data showed that the mitochondrial networks of cells were highly dynamic, continuously undergoing mitochondrial fission (fragmentation or division) and mitochondrial fusion (rejoining). However, at any given time, the majority of cancer cells had smaller, fragmented mitochondria when compared to healthy lung epithelial or vascular cells.

Working with a number of colleagues in our laboratories, we determined that lung cancer cells expressed higher levels of the mitochondrial fission protein Drp-1 when compared to multiple healthy cell types found in the lung. Inhibition of Drp-1 reversed the mitochondrial fragmentation and restored the degree of mitochondrial networking in malignant cancer cells to the levels we observed in healthy non-malignant cells. Similarly, over- expression of the mitochondrial fusion mediator Mitofusin-2 (Mfn-2) also increased mitochondrial networking. Importantly, inhibiting mitochondrial fission resulted in a cell cycle arrest of cancer cells and markedly reduced cancer cell proliferation. In vivo experiments using a tumor xenotransplant model showed a marked reduction in tumor progression when tumors were either treated with a pharmacological inhibitor of Drp-1 or when Mfn-2 was over-expressed.

Our findings are consistent with the observation that mitochondria undergo a cycle of fission and fusion which is coordinated with the cycle of cell division (mitosis). Our experiments suggest that targeting the mitotic fission of mitochondria may be a complementary approach to halt cancer cell proliferation. When we examined the tumor tissues of lung cancer patients, we found that tumor regions indeed expressed markedly higher levels of Drp-1 than healthy lung tissues. Whether Drp-1 levels are also higher in other forms of cancer and whether targeting mitochondrial fission in these other tumor tissues would be equally beneficial still needs to be examined in future studies.

Prior studies have detailed the role of Drp-1 in non-malignant cells where the protein appears to play a role in cell death. Drp-1 induced mitochondrial fission is a characteristic of mitochondrial apoptosis and short-term inhibition of Drp-1 can actually prevent cell death. However, other studies have also linked Drp-1 activation and mitochondrial fission to cell proliferation to mitochondrial fission because the cell cycle regulator Cdk1/Cyclin B regulates the activity of the mitochondrial fission mediator Drp-1. This suggests that mitochondrial fission induced by Drp-1 has two very distinct and nearly contradictory roles: cell death and cell growth.

In the highly proliferative cancer cells that we studied, Drp-1 appeared to be primarily acting as a mediator of mitotic fission, but it is quite possible that in other cell types or settings, Drp-1 may be more closely tied to regulation of apoptotic fission. The fact that the same protein regulates seemingly opposite processes of apoptotic fission during cell death and mitotic fission during cell proliferation may seem surprising. However, it is also reminiscent of the fact that mitochondria themselves can have apparently contradictory roles in cells, acting both as metabolic powerhouses as well as initiators of cell death.

From an evolutionary and teleological standpoint, a cell undergoing division (mitosis) would want to coordinate this process with the dividing and distributing of its mitochondrial organelles. Intact mitochondrial networks are probably difficult to distribute to daughter cells, whereas smaller, fragmented (?fissioned?) mitochondria can be easily distributed. Our study suggests that a reverse signal also exists, by which halting the mitochondrial fission seems to also halt the progression of the cell cycle.

As with any research, our study also points towards many unanswered questions, some of them highlighting the importance of how the nucleus communicates with other organelles: what are the specific mechanisms by which preventing mitotic mitochondrial fission signals back to the nucleus and halts the progression of the cell cycle? How does the cell coordinate the dynamics of other organelles during cell cycle? Could the dynamics of other organelles also be therapeutically targeted in cancer cells?

I remember there were many times I felt that, looking for a loving relationship was a waste of time. After being hurt a few times, and crying and becoming a bit bitter in the process, I decided I would never let love play me for?a fool again. When the man who is now my husband came along, I was not about to show my soft sultry side. He had?to work hard to win me over. He would always say 'I'm fighting for you!' The fact that he wanted me that much was what I guess eventually melted my heart.


I'm sure all of us have had our times when relationships had banged us up and we said, 'No more!' Sometimes it was just then, that?LOVE felt we were ready to have that special someone walk into our lives. I had to make a choice when that opportunity came along. Just go to any of the online tax prep sites and file a form 4868th That will give you a six-month extension for filing your return.However, if you owe any taxes (and it seems like you probably will), the extension does NOT apply to the money you owe. You need to pay that as soon as possible. Superintendent Recertification gets an Update : TILT-UP TODAY

MT. VERNON, IOWA (March 27, 2012) ? The Tilt-Up Concrete Association (TCA) ? a non-profit international organization that serves to expand and improve the use of site-cast Tilt-Up as the preferred construction method ? has announced an update to the recertification process for the Tilt-Up Supervisor and Technician Certification program ? a collaborative effort between the American Concrete Institute (ACI) and TCA.

At the recently held ACI committee meetings in Dallas, Texas, ACI?s Certification Programs Committee (CPC) approved an alternative re-certification for the ACI/TCA Tilt-Up Supervisor and Technician Certification Program co-sponsored by ACI?s Committee C-650 (Tilt-Up Constructor Certification) and the TCA. The update will allow supervisors and technicians to recertify based on completing continuing education hours instead of re-testing. Currently, individuals have to recertify every five years by successful completion of the current written exam.

?Several committee members felt that recertification by simply taking basically the same exam was a disservice to the program,? said Ed Sauter, C-650 committee member and executive director of the TCA.? ?Retaking the same exam doesn?t ensure that the candidate has continued to learn or grow as a professional in the Tilt-Up industry or that they are up-to-date on current practices. The modifications require evidence of continuing education and work experience.?

Candidates seeking recertification now have an alternative method that requires them to register 10 hours of approved continuing education during the 5-year period of their certification and provide verified work experience. The classes that apply will be pre-approved by the TCA and the Committee from offerings at such venues as the World of Concrete and the TCA Annual Convention that are relevant to a supervisorial position.? Pre-approved courses will be listed on the TCA website and, where possible, in programs and other promotional materials at the specific events. The requirement for verified work experience consists of a minimum of three Tilt-Up projects or on project with at least 100 Tilt-Up panels during the period.

For the foreseeable future, taking and passing the then-current examination will continue to be an option although the C-650 Committee does hope to transition recertification fully to CEUs and work experience after a suitable transition period. Technicians will be able to begin recertifying by complying with the continuing education requirements offered at the TCA Annual Convention this fall or by continuing to retake the exam.

The exam itself will be undergoing modifications over the next couple of years as the references are updated to the latest edition of the ACI-551 Guide and the new Tilt-Up Construction Manual (scheduled for release at the end of the year). Since all questions must be referenced, new and updated information means new questions and answers.? This will also impact the Spanish version of the exam and reference materials.

The TCA Annual Convention, to be held in Amelia Island, Fla. from Oct. 2-4, will feature a new Field Supervisor track that will provide enough hours in one event to fulfill the continuing education requirements for recertification.

BALTIMORE, Md. Today, the Interactive Autism Network (IAN), http://www.ianproject.org, the nation's largest online autism research initiative and a project of the Kennedy Krieger Institute, reports preliminary results of the first national survey to examine the impact of bullying on children with autism spectrum disorders (ASD). The results show that 63 percent of children with ASD have been bullied at some point in their lives. These children, who are sometimes intentionally "triggered" into meltdowns or aggressive outbursts by peers, are bullied three times more frequently than their siblings who do not have ASD.

"These survey results show the urgent need to increase awareness, influence school policies and provide families and children with effective strategies for dealing with bullying," said Dr. Paul Law, director of the IAN Project at the Kennedy Krieger Institute. "We hope that this research will aid efforts to combat bullying by helping parents, policymakers and educators understand the extent of this problem in the autism community and be prepared to intervene." Nearly 1,200 parents of children with ASD completed the survey. Findings show that these children (ages 6 to 15 years) are especially vulnerable to bullying, and point to a number of risk factors.

Where and When Bullying Occurs

  • While bullying occurred at every grade level, between 5th and 8th grades appeared to be the worst, with 42 to 49 percent of children with ASD in those grades currently bullied.
  • Children with ASD attending regular public schools are bullied at a rate of nearly 50 percent more than children in private school or special education settings.
  • Types of bullying most often reported include being teased, picked on or made fun of (73 percent); being ignored or left out of things on purpose (51 percent); being called bad names (47 percent); and being pushed, shoved, hit, slapped or kicked (nearly 30 percent).

Potential Risk Factors

  • While parents reported that 39 percent of children with ASD were bullied in the month prior to the survey, only 12 percent of their typically developing siblings, ages 6 to 15, were bullied in the same timeframe, indicating children with ASD are bullied at a rate more than three times higher than their unaffected siblings.
  • Across ASD diagnoses, 61 percent of children with Asperger's syndrome experienced bullying, a rate nearly double that of children with other diagnoses on the autism spectrum. This may be due in part to different school placement across the groups.
  • Behaviors and traits associated with becoming a target of bullying include clumsiness, poor hygiene, rigid rule keeping, talking obsessively about a favorite topic, frequent meltdowns and inflexibility.
  • Of those children who want to interact with others, but have a hard time making friends, 57 percent are bullied, compared to only 25 percent of children who prefer to play alone and 34 percent of children who will play, but only if approached.

Experience as Bullies and "Bully-Victims"

While children with ASD are frequently victims, they may also behave as bullies, or at least be viewed as a bully.

  • 46 percent of children with ASD have been a victim of bullying only, while 17 percent of children with ASD have been a bully-victim, defined as a child who has been bullied and also bullied others.
  • 52 percent of parents indicated that peers taunted their child to intentionally trigger a meltdown or aggressive outburst.

Researchers believe that the deficits in social understanding common in children with ASD may lead to bullying behavior by the child that is different than that displayed by typically developing children. For example, an honest but socially unacceptable remark such as, "You're fat," by the child with ASD may be viewed by others as purposely cruel when it is not. Likewise, a child with ASD who is accidentally bumped into might misinterpret this as intentional, and lash out in a way that looks like bullying.

"Children with ASD are already vulnerable. To experience teasing, taunts, ostracism or other forms of spite may make a child who was already struggling to cope become completely unable to function," said Dr. Law. "The issue is complex and we plan to carefully analyze the data and publish peer-reviewed findings that will serve to advance policy and care for individuals with ASD."

Uganda's new slave trade: Sex trafficking

Sitting at a table in a run-down bar on the edge of Ugandan capital Kampala, Stella Kobusingye stares into the distance as she recalls the tangle of lies and deceit that led to her sex-slavery ordeal.

Struggling to support her young son and ill mother with the income from a small shop, last year Kobusingye heard about a job opportunity earning $800 a month working at a Ugandan-owned boutique in Malaysia.

After triple-checking that the job was genuine, Kobusingye sold her shop, paid for a passport and bought a visa for China, where middlemen told her she needed transit, before being handed a ticket and boarding a plane.

But when she arrived in China, she was met by a Ugandan woman, taken to a hotel where scores of other Ugandan girls were staying and told she now owed $7,000 for the airfare.

"There were no jobs there, they had lied to me -- the only thing they had taken me there for was prostitution," said Kobusingye, who asked that her name be changed.

That night, a Nigerian man knocked on the door and said he had paid to have sex with her. Kept prisoner in her hotel room and with her passport taken away, for the next few weeks Kobusingye was forced -- sometimes brutally -- to sleep with up to five Nigerian men each night.

To scare her into silence, Kobusingye says her captors took her to a Ugandan witchdoctor there who used her fingernails and pubic hair to perform rituals.

Kobusingye's story is typical of a growing number of young and vulnerable Ugandan women who officials say are being duped into travelling abroad -- particularly to countries in the Far East and Middle East -- and then forced into prostitution.

In a country with massive unemployment and few opportunities, posters in shopping malls around Kampala advertise opportunities for well-paying jobs or studies abroad.

It is almost impossible to say how many Ugandan girls have been trafficked abroad but officials estimate in Malaysia alone -- a country with visa-free travel for Ugandans and growing economic ties -- there are currently about 600 Ugandan women in forced prostitution. No figures are available for the number of trafficked women in China.

During a recent visit to Kampala, Hajah Noraihan, a Malaysian working as Uganda's honorary consul in Kuala Lumpur, flicked through a series of horrifying pictures to show what happens to the girls who refuse to comply -- the face of a young woman thrown from a third-floor balcony, the body of another brutally murdered on a bedroom floor.

In the past two years, at least three Ugandan girls have been killed in Malaysia, she said.

Since late last year, Malaysian police have cracked down on the prostitution rings. In a single raid in October, 21 Ugandan women were discovered at just one location and there are around 60 currently being detained by Malaysian authorities.

With the help of the International Organisation for Migration, in the past five months 14 girls have been brought back to Uganda, Noraihan says.

Now Noraihan, a native of Kuala Lumpur, is pressing authorities in Uganda to deal with the problem at its source.

"We really need to see action on this, not just talk, not just reports. We need to see action on it and now," she said.

That pressure seems to be paying off.

Although several of those suspected of trafficking girls to Malaysia have already been arrested, critics say authorities have been sluggish or too poorly funded and trained to effectively crack down on those behind the rings.

Now though, officials from the immigration department to law enforcement agencies and presidential advisers have started meeting to discuss the issue and a group of lawmakers is pushing to visit Malaysia to undertake further investigations.

But public awareness of the threat remains low and the trafficking rings are highly organised and increasingly sophisticated, said Asan Kasingye, director of the Interpol office in Kampala.

"The criminals, these international rings, they are working against us. Instead of taking 10 girls, they might take one a week and if you go to the airport and there is a person going to Malaysia she will have documents saying she is going for studies," Kasingye said. And Uganda is just one of many sub-Saharan countries affected by sex trafficking.

Trafficking victim Kobusingye may finally be back home but she will never escape the effects of what happened to her.

After she attempted to find a way to flee China, Kobusingye's captors sent her to Malaysia, where they thought it would be easier to keep her.

But at Kuala Lumpur airport, immigration officials stopped Kobusingye. During several months in detention there she had a miscarriage.

Then she was told she had been infected with HIV.

Kobusingye was eventually sent back to Uganda with assistance from the International Organisation for Migration and the Malaysian consul. Hemsworth: 'Hunger Games' violence not gratuitous

LOS ANGELES (AP) ? "The Hunger Games" may be about kids who are forced to fight to the death on live TV, but one of its stars, Liam Hemsworth, said the violence in the film isn't for nothing.

"None of it is glorified and it's not about that," said the 22-year-old Australian.

"It's about kids in a horrible situation and there's this girl who overcomes it and gives hope to everyone and they come together to do something about it," he said.

In the PG-13 movie, which opens Friday, Hemsworth plays Gale Hawthorne, the best friend of Katniss Everdeen, portrayed by Jennifer Lawrence. His role in the first "Hunger Games" story is a small one but is more prevalent in the two stories that follow. (They're based on a best-selling book trilogy by Suzanne Collins.)

Hemsworth doesn't have any fight scenes in the first film but did have to lose "quite a bit of weight" to play a teen living in a poor area who hunts to help feed his family.

He recalled one memorable day off from dieting during filming in North Carolina when he and costar Josh Hutcherson visited a certain fast food chain.

"Josh and I went to Kentucky ? he's from Kentucky," Hemsworth said. "We decided to get White Castle, which I'd never had before, and then we played basketball after eating the White Castle and we threw up a little bit so that was interesting."

"The Hunger Games" also stars Woody Harrelson, Elizabeth Banks, Lenny Kravitz and Donald Sutherland.
